實驗步驟
1．首先將內層滅菌桶取出，再向外層鍋內加入適量的水，使水面與三角擱架相平為宜。
2．放回滅菌桶，并裝入待滅菌物品。注意不要裝得太擠，以免防礙蒸汽流通而影響滅菌效果。三角燒瓶與試管口端均不要與桶壁接觸，以免冷凝水淋濕包口的紙而透入棉塞。
3．加蓋，并將蓋上的排氣軟管插入內層滅菌桶的排氣槽內。再以兩兩對稱的方式同時旋緊相對的兩個螺栓，使螺栓松緊*，勿使漏氣。
4．用電爐或煤氣加熱，并同時打開排氣閥，使水沸騰以排除鍋內的冷空氣。待冷空氣*排盡后，關上排氣閥，讓鍋內的溫度隨蒸汽壓力增加而逐漸上升。當鍋內壓力升到所需壓力時，控制熱源，維持壓力至所需時間。本實驗用 1．05kg/cm2，121．3℃，20分鐘滅菌。
5．滅菌所需時間到后，切斷電源或關閉煤氣，讓滅菌鍋內溫度自然下降，當壓力表的壓力降至0時，打開排氣閥，旋松螺栓，打開蓋子，取出滅菌物品。如果壓力未降到0時，打開排氣閥，就會因鍋內壓力突然下降，使容器內的培養(yǎng)基由于內外壓力不平衡而沖出燒瓶口或試管口，造成棉塞沾染培養(yǎng)基而發(fā)生污染。
6．將取出的滅菌培養(yǎng)基放入 37℃溫箱培養(yǎng)24小時，經檢查若無雜菌生長，即可待

Experimental steps

After the cold air is compley drained off, close the exhaust valve so that the temperature inside the pan gradually increases with the increase of steam pressure. When the pressure in the pan rises to the desired pressure, the heat source is controlled and the pressure is maintained until the desired time. The experiment with 1.05kg / cm2, 121.3 ℃, 20 minutes sterilization.
5. Turn off the power or turn off the gas to allow the temperature in the autoclave to drop naturally. When the gauge pressure drops to 0, open the vent valve, loosen the screw, open the lid and remove the sterilized items.
