如何完成一次又經(jīng)濟的Tunel檢測？

——Yeasen TUNEL detection Kit 讓“美”原位呈現(xiàn)

檢測機理：

細胞凋亡晚期，染色體DNA雙鏈斷裂或單鏈斷裂產(chǎn)生大量的粘性3-OH末端，在脫氧核糖核苷酸末端轉移（TdT）的作用下，將熒光素/酶標記的dUTP結合到DNA的3-末端，從而可通過檢測熒光完成對細胞凋亡的檢測，這類方法稱為脫氧核糖核苷酸末端轉移酶介導的缺口末端標記法（Terminal-Deoxynucleotidyl Transferase Mediated Nick End Labeling,TUNEL），原理見圖1。

常見問題：

Q:出現(xiàn)非特異性熒光標記？

1）組織/細胞本身核酶或聚合酶活性水平較高，易導致出現(xiàn)非特異性的熒光標記，例如平滑肌細胞。解決方法是，取細胞或組織后立即固定并且要充分固定，以阻止這些酶導致假陽性。

2）使用了不適當?shù)墓潭ㄒ海缫恍┧嵝怨潭ㄒ?，導致出現(xiàn)假陽性。建議采用新鮮配置的4%中性對聚甲醛固定液。

3）TUNEL檢測反應時間過長，細胞或組織表面不能保持濕潤，也可能出現(xiàn)非特異性熒光。注意控制反應時間，并確保TUNEL檢測反應液能很好地覆蓋樣品。

Q:熒光背景很高？

1）高速分裂和增殖的細胞，轉錄水平高，有時也會出現(xiàn)細胞核中的DNA斷裂。

2）在激發(fā)光下長時間的暴露，導致假陽性的出現(xiàn)。

Q:標記效率低？

1）使用乙醇或甲醇固定會導致標記的效率較低。

2）固定時間過長，導致交聯(lián)程度過高。此時宜減少固定時間。

3）組織切片過厚，使得固定效果不理想，控制在10 μm以內(nèi)。

4）熒光淬滅。Fluorescence在普通光照10min就會嚴重淬滅。解決方法是需注意避光操作。
