介紹

免疫測(cè)定是一種生化測(cè)試，用于測(cè)量生物體液中蛋白質(zhì)的濃度。該檢測(cè)利用了抗體與其抗原的特異性結(jié)合。

有不同類型的免疫測(cè)定，該協(xié)議包含有關(guān)夾心ELISA（酶聯(lián)免疫吸附測(cè)定）的信息。在三明治免疫測(cè)定中，也稱為“非競爭性 ELISA"，抗原與抗體位點(diǎn)結(jié)合，標(biāo)記抗體與抗原結(jié)合。

測(cè)量位點(diǎn)上的抗原量。非競爭性 ELISA 方法的結(jié)果將與抗原的濃度成正比。這是因?yàn)槿绻粗獦悠分胁淮嬖诳乖?，?biāo)記抗體將不會(huì)結(jié)合。

程序

在測(cè)定之前，兩種抗體制劑都應(yīng)純化且不含白蛋白。檢測(cè)抗體必須貼標(biāo)。

11. 用粘性塑料蓋住板，在室溫或 -20°C 下孵育 60-90 分鐘。

用洗滌液洗滌四次。

與二抗（或生物素化一抗的鏈霉親和素-HRP）孵育

13. 加入 100 μl 標(biāo)記的二抗。要添加的量可以在初步實(shí)驗(yàn)中確定。為了準(zhǔn)確定量，應(yīng)過量使用標(biāo)記的二抗。所有稀釋都應(yīng)在稀釋緩沖液中進(jìn)行。

14. 用粘性塑料蓋住板，在室溫或 -20°C 下孵育 60-90 分鐘。

16. 按照制造商的指示添加基材。

17. 在建議的孵育時(shí)間過后，向每個(gè)孔中加入終止溶液。

18.添加終止液后30分鐘內(nèi)，可在ELISA讀數(shù)儀上測(cè)量目標(biāo)波長的光密度。

19. 計(jì)算每組重復(fù)標(biāo)準(zhǔn)品、樣品和對(duì)照品的平均吸光度值。

20. 如果單個(gè)吸光度值與相應(yīng)的平均值相差超過15%，則認(rèn)為結(jié)果可疑，應(yīng)重新測(cè)定樣品。

21. 使用能夠生成良好曲線擬合的計(jì)算機(jī)軟件減少數(shù)據(jù)，從而創(chuàng)建標(biāo)準(zhǔn)曲線。

22. 如果樣品已被稀釋，則根據(jù)標(biāo)準(zhǔn)曲線測(cè)定的濃度必須乘以稀釋因子。
